The theatre is under the ownership of the Nederlander Organization and is operated by the same. William Neil Smith does the architecture of the building. Nederlander Theatre was formerly named the Billy Rose Theatre and National Theatre. It is said to be one of the Nederlander Organization's nine Broadway theaters. The venue has been named for 20th-century American theater impresario entitled David Tobias Nederlander since 1980.
Walter C. Jordan built Nederlander Theatre for $950,000. Until 1956, the venue was owned by the Shubert Organization. In 1979, the Nederlander Organization purchased the venue and named it the Trafalgar Theatre. The venue is now known for hosting various shows, such as Edmond Rostand's Cyrano de Bergerac, Edward Albee's Who's Afraid of Virginia Woolf?, Noël Coward's Private Lives, William Shakespeare's Julius Caesar, and the Tony award-winning Rent.

Motown: The Musical, 2017: War Paint, 2018: Pretty Woman: The Musical, 2019: Kristin Chenoweth: For The Girls, Harry Connick Jr.: A Celebration of Cole Porter, and 2021: MJ: The Michael Jackson Musical. A box office record was achieved by Newsies when the production grossed $1,024,516.60 for eight performances; however, Pretty Woman: The Musical has the current gross record.
